Tissue Section Cutting
Cut the unstained slides at appropriate microns according to the stain ordered or tissue type.
Tissue section cutting. Also check the cryostat temperature. Sectioning chill paraffin embedded tissue blocks on ice before sectioning. In general a slow uniform cutting stroke is best.
If you are new to sectioning it is advisable that you spend some time developing your sectioning rhythm on practice blocks before attempting to cut valuable tissue. Do not ever stop and start a cut mid way through a section. The tissue is cracking in my sections put a warm finger on the block then try again.
The fatty tissue is cut without interfering with the tumor tissue and the margin is best preserved in this vertical section. Steel blades are used to prepare sections of animal or plant tissues for light microscopy histology. Place the blade in the holder ensure it is secure and set the clearance angle.
Important in science microtomes are used in microscopy allowing for the preparation of samples for observation under transmitted light or electron radiation. These sections are called frozen sections. Fill a waterbath with ultrapure water and heat to 40 45 o c.
The clearance angle prevents. Alternatively we can infiltrate our tissue specimen with a liquid agent that can subsequently be converted into a solid that has appropriate physical properties which will allow thin sections to be cut from it. Microtomes use steel glass or diamond blades depending upon the specimen being sliced and the desired thickness of the sections being cut.
